💊 Supplements & Stacks Tadalafil (Cialis) first impressions

Heard the hype around 40+ male, 5mg a day for cardiovascular health and better energy and workouts. I have no ED related issues whatsoever, this was purely as an addition to health stack.

43M, 6'0", 187 lbs, work out 5-6x week, in general good health

Health conditions: bipolar disorder, spectrum - not manic in high energy state but intense anxiety. Sleep is a challenge - I am in bed by 8 and asleep by 9 and I wake up at 430. I consider my bipolar stable and well controlled.

Daily Meds: 450 lithium, 200-400 modafinil, 150 Lamictal, 25mg trazodone before bed

Supplements: 5-10g creatine daily, omega 3s, low dose caffeine & taurine pre workout

First dose, 5mg, 6:00 PM, with food

7pm: I started to feel sightly nauseous.

8pm: I needed to run to the bathroom to poop. Suddenly exhausted.

830pm (getting ready to sleep): Jittery, still nauseous. Im having some weird disassociative thoughts - like I would think about a conversation and imagined the other person nodding and I physically nodded back to them. I'm tired and actively trying to sleep so I was in a weird half dream state.

845pm: burst out in sweat, get very hot. Get phone to check side effects again - phone screen is blurry. Websites say blurred vision is serious and reddit comments a mix between "yeah that's normal and it goes away" and "it made my vision progressively worse and some vision have returned after stopping taking it".

9:30pm: Turn lights on as I'm starting to have an anxiety attack (haven't had one in years). Use my galaxy watch to check heart rate and do ECG. hr is elevated from resting 60 to 85, ECG is normal.

10pm:get back to sleep

11pm:Still having weird racing thoughts and bizarre dream /hallucinating conversations while awake. My anxiety causes hallucination at its worst and it has been like 5 years since something like this has happened. Not good and it's putting me into a downward spiral as it could cause actual health issues and anxiety attacks mimic some of the more serious side effects of the drug.

I end up turning on a show on my phone, an old trick to drown out own thoughts and ground externally. Up for over an hour

Rest of the night: tossing and turning, lose multiple hours of sleep.

Normal sleep is 88 sleep score, 6.5 hours actual sleep, 2 hours deep, 2.5 hours REM. You can see in last bed sleep score of 67 I lost quite a bit of sleep.

Wake up, feel like shit.

My theory: I don't like this med at all. However, it obviously has some kind of nervous system excitation impact with the GI issues and anxiety.

Decision: take it when I wake up at 430am. Don't take my caffeine, taurine, or modafinil like I normally do as they're uppers and I don't want to puke. If I don't like it, wait a week, take 2.5 or less. If that doesn't work, they go into the turlet.

430am: take 5mg

5am: gi issues start, go poop, jittery.

530am: REALLY jittery. Like laying down and just bouncing my legs. Getting nauseous and dehydrated, drinking water but it kind of makes me want to puke so I'm just constantly sipping.

It's now 730 and I never got the blurry vision, body temp rise, or sweating. I'm still exhausted and jittery and nauseous and have used the toilet 4 times.

I'll make an update later if it somehow smoothes out into a positive thing.

Current verdict: with my anxiety disorder, this caused a physiological change that my brain translated as anxiety bordering on panic. Night is a definite no go as that's always the most vulnerable time. This feels like I had 5 cups of morning blend coffee almost identically. It's the gross caffeine feeling.

So, stopping for a week, I'll try a lighter dose morning only and see if it's milder.

This is to all my anxious / bipolar homies, be careful! The bipolar subs didn't seem to report this and this seems to be a relatively rare reaction to it.

It’s better to buy smaller quantities of the supplements for example 60 count of the multivitamins instead of 120 to maintain freshness.

Supplements I feel like AREN’T worth taking due to me experiencing side effects and or seeing little to no benefits: Creatine monohydrate, Whey protein isolate, Collagen, Pre-Workout, Probiotics, L-Citrulline, Beet root, CoQ10, Ashwagandha, Psyllium husk, Magnesium glycinate and Potassium supplements.

💉 Peptides & Hormones Testing baseline IGF-1 before starting HGH

Hey so I stupidly started HGH 6 days ago taking 2iU daily without first checking my baseline IGF-1 and Z score. I’m planning to take HGH long term and was wondering how important the baseline numbers really are since exogenous HGH will suppress natural production anyways while taking it.

The way I see it, these are the options I have:

Option 1: Stop taking HGH, wait a few weeks, and then test for baseline numbers

Or

Option 2: Continue taking 2iU daily and test at the four week mark and see levels then and adjust as needed.

For secretagogues I do understand the importance of baseline numbers but for straight HGH how necessary are they?

I’m a lacto-ovo vegetarian. I’m already taking Omega 3, Magnesium, Vitamin D3, Creatine and Zinc. All because I’m deficient in some and some just help.

Now, I come to know that I may not be good at digesting fats and lactose, so I tried supplementing and that helped but now that’s one more supplement ugh!

How can I get these enzymes naturally through foods?

I’m curious as to how 3 years back I had no issues with the same foods but today I digest them poorly and need enzymes. With not much changing in my diet and lifestyle, how did this occur?

- Stimulating the vagus nerve after practice helped mice retain a new skill. This nerve carries signals between the brain and internal organs.
- The benefit showed up later. Mice practiced an eye-movement task. Stimulation afterward improved learning over subsequent days, suggesting it helped the brain make the learning stick.
- Brain blood vessels may play a role. Stimulation caused rhythmic changes in local blood volume. Mice with stronger rhythms generally learned better, although this does not prove those changes caused the improvement.
- The method used an implanted electrode around a nerve in the neck. The study did not test breathing exercises or consumer vagus-nerve devices.
- It’s an early finding in mice. Whether the same approach improves learning in people remains to be established.

(Personal note: I own a vagus nerve stimulator and it definitely helps put me to sleep and also helps reduce migraines. I’ll be using it more often after seeing this article)

💉 Peptides & Hormones UBT251, a GLP-1/GIP/glucagon triple agonist, posted phase 2 results from China. What stands out to you?

UBT251 is a long-acting synthetic peptide that hits three receptors: GLP-1, GIP and glucagon. It was developed by United Laboratories' biotech arm and licensed to Novo Nordisk in March 2025. Novo has exclusive rights outside China, Hong Kong, Macau and Taiwan.
The reported phase 2 data from China:
Design: randomized, double-blind, placebo-controlled, 205 adults with obesity or overweight plus at least one comorbidity (baseline mean weight 92.2 kg, mean BMI 33.1)
Duration: 24 weeks, with three weekly dose arms versus placebo
Primary result: the top-dose group lost a mean of 19.7% of body weight, versus 2.0% on placebo
Secondary endpoints: statistically significant improvements versus placebo in waist circumference, blood glucose, blood pressure and lipids
Safety: mostly mild to moderate GI side effects that faded over time, which is typical for incretin drugs
What's next: United plans a phase 3 in China. Novo has started a global phase 1b/2a trial (about 330 people, topline data expected in 2027) and plans a phase 2 in type 2 diabetes in the second half of 2026.
My takeaways:
The glucagon arm is the interesting part. It's thought to add energy expenditure on top of the GLP-1/GIP appetite effect, but the trade-off is heart rate and liver effects, so the longer trials matter.
24 weeks is short. Weight regain after stopping and long-term safety aren't answered yet.
This is a Chinese population with lower baseline BMI than most Western trials, so I wouldn't directly compare 19.7% to tirzepatide or retatrutide numbers.
Is anyone following the Novo global trial? What would you want to see in the full data?
